FOXO4-DRI (also known as FOXO4-D-Retro-Inverso, Proxofim, or FOXO4-DRI peptide) is a synthetic cell-permeable retro-inverso peptide composed entirely of D-amino acids. It is designed as a competitive antagonist that mimics the p53-binding domain of the FOXO4 (Forkhead box O4) transcription factor, thereby selectively disrupting the FOXO4-p53 interaction. This research compound has attracted substantial scientific interest as a prototype senolytic agent that preferentially induces apoptosis in senescent cells while largely sparing healthy proliferating cells. Preliminary and advanced preclinical studies demonstrate that FOXO4-DRI promotes nuclear exclusion of p53 in senescent cells, redirects it toward mitochondrial pathways, triggers cytochrome c release and caspase activation, and ultimately clears senescent populations. Researchers utilize FOXO4-DRI in laboratory and animal models to investigate mechanisms of cellular senescence, tissue homeostasis restoration, age-related pathologies, chemotherapy-induced toxicity, and regenerative interventions. As a research-grade compound, FOXO4-DRI is intended exclusively for in vitro and animal studies. Scientists value its high selectivity, enhanced proteolytic stability due to the D-amino acid and retro-inverso design, and its role in advancing understanding of FOXO4-p53 axis-mediated senescence survival.

01 Selective Clearance of Senescent Cells
Preclinical studies show FOXO4-DRI potently and selectively induces apoptosis in senescent fibroblasts, chondrocytes, endothelial cells, Leydig cells, and other cell types by disrupting FOXO4-p53 complexes in PML bodies/DNA-SCARS, leading to p53 mitochondrial translocation and execution of the apoptotic program.
02 Reversal of Age-Related Tissue Dysfunction
In naturally aged and accelerated-aging mouse models, FOXO4-DRI restores multiple fitness markers (including renal function, fur density, physical performance, and overall tissue homeostasis) by eliminating accumulated senescent cells and reducing the senescence-associated secretory phenotype (SASP).
03 Chemotherapy-Induced Toxicity Mitigation
Investigations reveal that FOXO4-DRI alleviates doxorubicin-induced chemotoxicity by targeting senescent cells in affected tissues (e.g., kidney, liver), reducing inflammation and helping restore organ function after chemotherapeutic stress.
04 Reproductive and Endocrine Health
Research demonstrates benefits in age-related testosterone insufficiency models by selectively removing senescent Leydig cells in the testes, improving the testicular microenvironment, spermatogenesis, and hormone secretion in aged mice.
05 Broader Regenerative and Disease Models
Emerging studies explore applications in models of vascular aging (endothelial senescence), keloid formation, osteoarthritis (chondrocyte senescence), and other senescence-driven pathologies, highlighting its potential to enhance tissue regeneration and counteract chronic inflammation.
