N-Acetyl Semax Amidate (also known as N-Acetyl Semax NH₂ or Ac-Semax Amidate) is a synthetic heptapeptide derived from Semax with two key stabilizing modifications: N-terminal acetylation and C-terminal amidation. These changes significantly improve enzymatic resistance (especially against aminopeptidases), enhance metabolic stability, and often improve blood-brain barrier penetration and duration of action compared to standard Semax.
This research compound retains the core ACTH(4-10) analog sequence (Met-Glu-His-Phe-Pro-Gly-Pro) and is studied for its potent nootropic, neuroprotective, neurotrophic, and cognitive-enhancing properties. Preclinical research suggests it strongly upregulates BDNF and TrkB expression, modulates dopaminergic and serotonergic systems, reduces oxidative stress and neuroinflammation, and supports neuroplasticity with potentially superior pharmacokinetics to unmodified Semax.
Research Applications
-
Cognitive Enhancement & Nootropic EffectsStudied for improvements in memory, learning, attention, and mental performance under stress, fatigue, or aging conditions.
-
NeuroprotectionInvestigated in models of cerebral ischemia, stroke recovery, traumatic brain injury, and neurodegeneration for its ability to reduce neuronal damage, apoptosis, and oxidative stress.
-
BDNF & Neurotrophic SupportPromotes BDNF expression and hippocampal neurogenesis, supporting long-term synaptic plasticity and cognitive resilience.
-
Anti-Inflammatory & Anti-Stress ActionModulates neuroinflammation and helps normalize neurotransmitter balance during acute or chronic stress.
-
Peptide OptimizationThe dual modification (N-acetylation + C-amidation) is commonly researched for better stability, longer half-life, and enhanced intranasal or systemic efficacy compared to native Semax.
Storage & Handling
Storage & Handling
Lyophilised vials are stable at −20°C for up to 2 years unopened. Once reconstituted, refrigerate at 2–8°C and use within 28 days. Avoid repeated freeze-thaw cycles.
Beyond Use Date
Pre-filled formats (cartridge, pen, nasal) carry a beyond-use date of typically 4–8 weeks from receipt. ≥98% purity by HPLC. For in vitro and preclinical research use only — not for human or veterinary use.
